1.1 Project Background
1.1.1 P roduct Introduction
Pulullan is a kind of extracellular water-soluble viscous polysaccharide fermented by aureobasidium pullulans fermentation , similar to glucan and xanthan gum, which is a special microbial polysaccharide discovered by R. Bauer in 1938 . This polysaccharide is a straight-chain polysaccharide polymerized by maltotriose repeating units connected by α-1, 4 glucosidic bond via α-1 , 6 glucosidic bond , with molecular weight 2 0,000 - 2 million and degree of polymerization 100 - 5,000 . (General molecular weight of commodity is about 20 0,000 , consisted of about 480 maltotrioses) . This polysaccharide has two important features: elastic structure and big solubility . Due to excellent features of strong film-forming property, gas barrier property, plasticity and viscosity, and easily soluble in water property, zero poison or harm, pulullan has been widely used in medicine , food, light industry , chemical and petroleum fields . On May 19, 2006, the Ministry of Health published No. 8 announcement , pulullan was one of four newly added food additives , and can be used as coating agent and thickener in candy, chocolate coating, diaphragm, compound seasoner and fruit and vegetable juice .
Pulullan is a substance with high resistance to the stress state formed due to environmental change , and a typical emergency metabolite in the organism. Lots of researches indicate that, stress tolerance of many organisms expressed in stress environments (such as hunger, high temperature, freezing, dryness, high permeability, radiation and toxic substance) is directly related to pulullan content in the body, because in various harsh environments , pulullan can show up good protection for the species biological film , protein and nucleic acid and other biomacromolecules .  
Pulullan can protect drug-carrying liposomes, prevent starch aging and protein denaturation, restrain grease substances from rancidity and fishy taste from generating, alleviate odd flavor of chicken, duck and goose, restrain rise bran smell, strengthen retaining freshness of vegetable , meat and fruit, stabilize superoxide dismutase in the materials, prevent tooth decay and supplement energy . In addition, pulullan has certain influence on the stress resistance of plant , for example, research of salt tolerance, cold resistance, drought resistance and water stress resistance of exogenous pulullan on cucumber indicates that, it has positive effect for stress resistance of plant . In short , exogenous pulullan has good non-specific protection function . Therefore , pulullan is also called “sugar of life” .  
1.1. 2 Market Prospects
According to the present production cost , the price of pulullan is about 5 0,000 -7 0,000 yuan/ t, lower than the ma rket price 200,000 yuan/t . After cost reduction, application of pulullan in the industries out of capsule, such as cosmetic , food, chemical industry , environmental protection and pesticide , can be effectively driven. The market potential is enormous .
Pullulan has many excellent features and broad application prospect , attracting the attention of scholars. Only World Patents Index of November 1983 contained 152 latest applied patents in relation to aureobasidium pullulans . Japan conducted research for aureobasidium pullulans very early , and carried out production at the level of pilot plant test, thus forming large-batch production scale , which still has monopoly of international market ; America also bred excellent bacteria in the middle of 1970s, and has put into batch production; however, China conducted related research in the beginning of 19 80 s, and is still at the level of laboratory research , and has not seen the report of industrialization production . At present, different specifications of pullulan products in China are used , but they have not been put into production in large scale, and most of them are still in laboratory or pilot plant test stage . The existing application researches are mainly about it used as preservative of food ( e.g. egg, marine product ) and fruits ( e.g. apple, pear ).
So far , Japan Hayashibara Co., Ltd. is still the only manufacturer of pullulan , but China still does not produce this product. At present, China imports 200t from foreign countries ( mainly from Japan Hayashibara Co., Ltd. ) every year at expensive price 30 0,000 yuan / t. It is mainly used in cosmetic industry . With Ministry of Health 2006 No. 8 Announcement approving new varieties such as pullulan and other food additives , promotion and application of pullulan in food and medicine industries will continuously develop. The estimated annual demand of medicine industry is about 2000 t ( only capsule preparation . Western Europe is influenced by mad cow disease, and some Islamic states refuse using animal capsule , it will be inevitable of replacing animal capsule with plant capsule. As raw material of plant capsule, pullulan has unique advantage . China's annual consumption of gelatin used in capsule preparation is more than 10 , 000 t ) . The market prospect is optimistic .
1.1.3 Technical Analysis  
This project relies on the laboratory achievements constructed by Qingdao Institute of Bioenergy and Bioprocess Technology, CAS.
(1) Established methods for bacterial strain mutagenesis and screening , fermentation control , separation purification and product quality evaluation , and obtained core bacterial strain with low pigment and high output .
(2) Preliminary optimization of fermentation conditions obtained the optimum carbon source and important factors influencing output and growth , and laid foundation for the subsequent fermentation control .
(3) Established 5L level fermentation technology. At 5L fermentation tank level, realized output 109g/L and yield 40-45% (high er than existing output 60g/L ).
(4) Established separation and product quality inspection and evaluation method . Analyzed products a ccording to National Standard GB 28402-2012 for pulullan, and made the obtained products reach the national standard .
1.1.4 A dvantageous Conditions of Project Construction
( 1 ) Resource advantage
Jilin Province is a major province of corn industry, annual output of corn starch reached tens of millions of tons , and still constantly rising , but the market price changes substantially. Therefore , it is urgent to conduct deep processing of corn starch and increase market competitiveness of corn starch downstream products . This project is proposed and completed under this big background as key research project of Jilin Province . This project takes corn starch as raw material to develop corn deep processing product - pulullan with very high additional value and enormous market potential , the edible grade market price is about 8 0,000 yuan/t , medical grade over 30 0,000 yuan /t. Therefore, production of this product will double the corn value, and it will become true “yellow petroleum” and “soft gold” .

Gongzhuling City has convenient traffic conditions , Beijing-Harbin double-track railway, Harbin-Dalian Expressway and 102 National Road form northeast transport artery traversing Gongzhuling City . Harbin-Dalian Expressway has two exits at the Gongzhuling section . Gongzhuling City is only 50km from Changchun Civil Aviation Airport , 720km from Dalian Port, and 440km from Yingkou Port . It takes only half an hour to reach Changchun by car, 2 h to Shenyang, and 5 h to Dalian , and the whole journey is connected with expressway .

2.2 Overview
Gongzhuling Chemical Industrial Park was approved and established by the Provincial Department of Environmental Protection in April 2011, which is one of only three chemical industrial parks approved in Jilin Province.
